Every time i try to scan something on my new scanner, Canoscan 4400F, it says it cannot access TWAIN and to check my connection and try again. I have unplugged and re-plugged the scanner and restarted my computer. What now?

I had the same problem with a new canoscan 8800f. I found by connecting the scanner USB lead to a USB 1 .1 port instead of a USB 2.0 port the problem seems to have been was solved. I was fortunate to have both. Originally it was just USB 1.1 ports on my PC, but I added USB 2.0. I don't know how you solve the problem if you only have 2.0 ports.
Try going to CanoScan Toolbox > Settings and reset the 'defaults' button at bottom... this will restore factory settings and hopefully fix your problem... The other error message you can expect to get is "unable to scan - insufficent memory, check your **** space' (or something similar)... same fix works for both! ...Have had both messages on mine, sounds like a semi-regular conflict on some sort, but seems to be easily fixed. uninstall all drivers and software that cam with the scanner, and then try installing the software again before connection the scanner... this is a problem when there are two different twain drivers, look for C:\Windows\twain_32\CNQ4803(2) see if there is a driver in there.
