Unlike the Curious Village (the first game in the series), each version of Pandora's Box has a single language on the cartridge (so the Spanish version only has Spanish)
Operationally, there's really not much to know about the Professor Layton games and everything (such as tapping on the shoe to move, opening the trunk and saving) can be easily worked out
Saying that, there's little point in playing a Professor Layton game in a language you don't understand since you wouldn't know what's happening in the story (which is a big part of the game) and you also wouldn't be able to solve the vast majority of puzzles (since you wouldn't understand the questions, which are largely text based)
If you want to get through the Spanish version (to, say, watch some cut scenes and solve the few puzzles that would be understandable) then you can follow this fully illustrated step by step walkthrough

On a side note, Pandora's Box is the second game in the series and is best played after the Curious Village (as there's a sub-plot running through the games and the second game spoils one of the major plot twists in the first game)
