I would say that your "Output Bearing" has gone, this can be confirmed by removing the chrome cover covering the secondary and then the actual secondary cover and "Swinging" up/down side to side on the shaft that comes out of the engine. If there is any movement in the shaft, then that is the cause of the noise.
To repair this, the engine will have to come out and have the bottom of the engine removed to gain access to the bearing. A new (Better after market) bearing is available from www.cademaster.com under Cavalcade parts tab. Tracy in KS, Ed in St Louis or Jerry in Wi can do this for you if they are nearby to your location.

Suzuki intruders are shaft drive only unless someone has engineered a major conversion to belt drive
when refitting the rear wheel something has gone wrong and it may be binding on something and causing the noise remove rear wheel and check for points of contact if none shaft drive ,gearbox and clutch need to be investigated

the front drive axle on one side may have not been installed all the way in to the transmission, check to see if it is loose and have some one look under front end with engine on, front end jacked up, emergency brake on and in gear to see if a axle is turning. if one is and yhe other is not put the car in park and turn\ roll the wheel that was not turning by hand if it rolls your problem is most likley to be the axle has come loose from the transmission
