You have two options. If you normally transfer your photos by connecting your camera to your computer, remove the memory card before making the connection. This will allow the computer to see the photos in the camera's internal memory.
If you normally transfer your photos by putting the memory card into a card reader, use the Backup command in the Settings-1 menu to transfer the photos from the camera's internal memory to the memory card, then transfer the photos from the card to the computer as normal. With no card in the camera, go into the menu and find "format". Follow the on-screen instructions to erase the internal memory. After that, your card should work properly.

The best way to download pictures from your camera to your computer involves removing the memory card from the camera and plugging it into a card reader (either built-in to the computer or connected via USB or FireWire). This is likely to be faster than connecting the camera to the computer, and won't run down your camera's batteries.

Formatting in the camera should have worked. Make sure the slide switch on the side of the card is in the unlocked position. With most cameras, when you bring up a picture and press the delete button, it gives you the option to delete that one picture or all of them. Have you tried that?
