you have not said what book order.
but see if this link points you where you need to go. How do cancel book order Google Search To cancel an item or an entire order:
Go to Your Orders.
Click Cancel Items. Note: ...
Select the check box next to each item you wish to remove from the order. If you want to cancel the entire order, select all of the items.
It's instant, and as far as I know they can't tell if you sent it to your Kindle or not, you'll either have to buy it again or transfer the file once you download it.
It may be that you accidentally ordered it twice, and they've just canceled the second order. Check in your Amazon account (Your account-Your orders-Digital orders) and see if you've actually been charged for this book. If you have, and it's really not available for downloading to your Kindle or already on it, contact them through <https://www.amazon.com/gp/help/customer/contact-us> with the dates you ordered and paid for the book. I've always found Kindle customer service swift, competent, and fair in adjusting errors.
