Here's a link to the detailed instructions for this repair. It's not too difficult, just a little tedious dealing with small parts.
Good luck!

2001 Toyota Sienna r.h. sliding door ''rear latch switch'' location

To start, the noun "''rear latch switch'' is incorrect. It's actually called "Slide Door Lock". It's located on page BO-20 of the Toyota Repair Manual for the 2001 Sienna. There are two door locks on one sliding door. One in the front and one in the rear. Obviously, you want the one in the rear. The location for the "Slide Door Lock" (which also contains an electric switch) is in the rear of the sliding door located mid way up door. You can see (sliding door opened from the back) it without dissembling the door. And, the lock is made up of plastic (dirty white in color).

Rear door on 2001 sienna won't open

The door handle might be broken, you need a new part. I got mine online for $40. keywords: 2001 2002 2003 2004 2005 2006 2007 Toyota Sequoia Metal Lift Gate Exterior Door Handle for Rear Outside Van Tailgate Handle. You need to remove the back door panel to replace this.

Both manual sliding doors on my Toyota Sienna will not open

If you can remove the inner door panel you will see a maze of cables running to the various locking points on the sliding door. If the cable going to the FRONT latch on the sliding door is broken (or in my case, simply dislodged inside the door) the door acts as if it is frozen shut.

The front cable is short, and easily replaced even by a newbie mechanic.

Rear Door Handle Broken

I had the same problem, and here is the solution:
http://www.findonefindall.com/toyota-sienna/siennareardoor.htm

It includes step-by-step instructions and photos. I printed it and headed to the garage. I while later (hours actually) it was fixed. I did end up going to the hardware store and buying the EXACT size screw they recommend. I went with a screw rather than a nut and bolt. Worked fine.

2004 Sienna LE Rear door won't unlatch

Older Sienna door handles have been known to age prematurely, many plastic ones cracked and broke a few years down the line. For the back hatch. a cable is attached to a plastic piece and hooked through the latch handle. That cable might have come off of the plastic piece or the piece had simply broken in two. If you are lucky, you might be able to superglue the plastic piece back such that the cable would pull and open a few times before breaking down again - it is a quick and dirty fix. Get it repaired properly so you do not end up using the sliding door to access the middle row as storage compartment....
