Have you tried a quick reboot? Of course, save the file you want to print, reboot your PC and try printing it again. If this doesn't work, try removing your printer in the "Devices and Printers" that you can find in the control panel (or Printers in XP). After removing, try adding a new printer again. Don't worry, you won't need drivers then because the driver is already in your PC after you installed the printer the very first time.

Also make sure that you do not have "pending" print jobs, as this can "overload" or "confuse" the printer. Clear all print jobs (cancel) and then restart computer (save work first) and then try printing again. If this does not work, un-install the driver for the printer (if you have the installation disk), then reinstall the printer driver with the disc. This will refresh the printer installation state on the computer. You will want to unplug the printer, from both the computer and the wall outlet, and follow the installation instructions specifically.

I had the same problem with the Lexmark Z645 and Vista.
After a lot of effort I gave up and emailed Lexmark.
Their response was surprisingly helpful:
First download and run "CleanupUtility.exe" from: ftp://ftp.lexmark.com/driver/inkjet_drivers/Cleanup/
Then unplug the printer's USB cable and restart your PC.
Then download and run the printer's drivers/install program
for Vista, from: http://downloads.lexmark.com:/cgi-perl/downloads.cgi?ccs=229:1:0:548:0:0&emeaframe=&fileID=10374&searchLang=en&searchLang=en&os_group=Windows%20Vista
Only plug the USB cable back in when the drivers/install
program tells you to.
After install, I think it asks for another reboot.
Then all should work fine.

If you double click on the print que, look at the status heading...what does it say? If it says printing and then windows gives you a "this document failed to print", then you probably have a driver issue. Delete the printer and re-install. If it says paused then right click on the job and clickon resume. You might also want to check in the printers and faxes folder, right click on the driver for the printer, see if an option is there that says resume printing, click it if it does. Also right click on driver and select properties, under the ports tab see if the correct port is selected. Best I can do without knowing more detail.

Here are instructions on how to delete the print jobs stuck in the queue. Follow the instructions carefully:
First, you have to stop the Print Spooler. To do this:
1. Click on the Windows logo start button.
2. Go to Control Panel
3. Open Administrative Tools
4. Open Services
5. It will pop-up a Services window. Scroll through the list and look for Print Spooler.
6. Double-click on Print Spooler, make sure Startup type is Automatic and click on the Stop button. Click on Ok button. Set the window aside.
Second, you need to clear the print jobs in the Spool folder. To do this:
1. Click on the Windows logo start button.
2. Type Spool in the Search bar(Vista) or click on Run(XP) and type Spool
3. Open Printers folder
4. Delete all the files in the Printers folder. Close the window.
Third, you have to enable the Print Spooler. To do this:
1. Go back to Services window.
2. Double-click on Print Spooler, and click on the Start button. Click on Ok button. Close the Services window. Click on the Windows logo start button. Go to Control Panel and open Printers folder. Refresh the page. That should clear the print queue now. Try to print again. If problem is not resolved, maybe it's a communication problem. You probably need to reinstall software. provide the type of printer, make and model. Also the type of connection, usb, parallel, ethernet?
Also did you verify that the default printer driver is the designated one for the particular make and model of the printer you are printing to?
