are you on a desktop or laptop? if on a desktop sometimes there are multiple places to plug in your mic, make sure you have not only plugged it in the right port, AND checked to see how high your mic sensitivity is on the software side.
on skype, click TOOLS, OPTIONS, Audio settings and talk, if you see the green power meter lighting up then make sure to slide the sensitivity up, or check and uncheck the auto feature, depending on where you had it try this and see what you get. Also turn donw your speakers as if your speakers are getting a feedback loop or are hearing the other people talk louder than it can hear you then it wont work either.
Check your video settings within skype, make sure to do a test call with skype as well and make sure that you log onto microsoft and get the updates for the microsoft lifecam.
