Western Digital hard drive inside is fried, check their website for information, they replace it for free if the drive itself is still in warranty, it might be gave it a shot.

You have to install the backup software on the new computer. Either download from WD to use what came with the MyBook. Attach the MyBook and find it in My Computer. If it is not there, you have to reboot and find the attached device. After Windows recognized the drive, the backup software will ask you for a new backup but just decline and reactivate the plan (actions/reactivate). That should do it!
Your drive has failed. to try to get back some of your items put the drive in the freezer for a day. the chill will make the heads condense enough for about 3 hours of use for emergency backup.
Regretfully, Western Digital drives have been known to fail and the sign is a fail to recognize, and the infamous clicking. I've not found any solution other than attempt data recovery with Disk Warrior or something similar.. There are data recovery companies but they aren't cheap...
If all you want to do is transfer data that you have backed up on your main computer then all you need to do is plug the drive into another computer, click on 'my computer' and click on the mybook drive to explore the data on it. To transfer a certain file just copy it and paste it onto the drive of the new computer.
If you are talking about backing up data from a second PC, you will need to install the WD software on that particular PC, and use the advanced backup menu. Just remember that if you back up a second computer, you need to create a folder for that data to be stored in, otherwise it is possible that it would overwrite the data from your first PC
in otherwords to back up several PCs the root of the WD drive should have folders in it labeled - 'PC 1', 'PC 2', etc
I have 6 My Books I use for various types of data storage (2x 1Tb and 4x 500GB). I experienced problems with 2 of the 500GB drives over the last 3 months. Speaking with a tech at Best Buy, apparently the problems are common and are related to heat buildup inside the case.
I think the problem is not with the drive itself, but rather with the compartment (specifically with the USB or Firewire interface). The failures I experienced are intermittent: Windows does not recognize the drive and starts displaying an error about an "USB device not recognized". The first time this happened I let the drive sit and cool, and it worked again. I tested it for bad sectors and it came back healthy. Then after about a day of use, the same USB error hapepned and the drive became unreadable again. If you open the enclosure, remove the hard drive, buy a different enclosure in install it, I think it will work and you will get your data back!
Fortunately, I always do a back up a backup so I did not lose anything. Needless to say, however, after this kind of unreliable behavior, the drives are going out the door and I will replace them with a SCSI external disk enclosure. More expensive, but reliable. I will eventually ask WD to replace the bad ones, but I am not sure I will trust them.
As for NTFS vs FAT, you can re-initiliaze the drive with NTFS using DIsk Management without a problem. I did this to all of mine (usign WInXP Pro).
I have had this problem with several MyBook hard drives. They work fine for a bit and then the windows xp OS fails to recognize them. If they do show up the computer warns me they are not formatted. My solution does not help fix the MyBook as is. However, if you have data you must have on it this may be of help. I simply opened up the MyBook plastic case. It is different for each drive. The Pro/premiums I have found you must remove the rubber packing to get at the latch area. After I removed the bare drive from the holder I used an IDE/SATA adapter kit with external power and plugged directly into a USB port. The drive was recognized immediately and I offloaded the data. The drive actually works better now then when it was in the Mybook holder. I am taking no chances though and have switched to LaCie drives. Hope this helps somewhat.
