How do I adjust the mixer to remove humming noise?

I used to be a hi-fi enthusiast many years ago.
A low frequency hum can be caused by inadvertently creating a "hum loop". This is when audio separates are earthed or grounded where they are plugged into the power outlets and then connected together using screened audio leads where the screening is connected to both plugs.
The situation can become worse when one of the separates is a type that isn't grounded.

Ineffective or insufficient power supply smoothing and/or shielding leaving an AC ripple on the DC also creates a hum or in the case of poor design or bad shielding can radiate AC magnetism into the high gain input stages of amplification.

Whatever the cause of the hum it can be aggravated by poor matching of separates with regard to input/output sensitivities and impedances. If the pre-amp stage gains have to be high to compensate for a low input signal the signal-to-noise ratio will always be poor through every stage. Small signals requiring large amounts of amplification requires low impedance circuitry and appropriate sensitivity.

How to connect Samson410 to the audio 2000 Karoake/pa system?

I'm guessing a bit because I don't know exact model numbers, but heres my best guess.
Connect the "pre out" L and R from the Karoake system via 1/4" cables to the 410 unit to line 1 and 2. From there, operate the Sampson unit as if you had two instruments plugged in to it.

Trying to connect laptop and getting a hissing noise.

You have not told us how you are attempting to connect the laptop. You would likely use a 1/8 stereo to RCA Wye cable to plug into the CD input jacks. You could also have a 1/8 Wye stereo to 1/4 inch jacks and use two of your input channels. The computer and the mixer need to be powered from the same receptacle. The sound boards in laptops sometimes are noisy. If the above doesn't fix it, buy a computer audio interface such as a UA25EX or others. Some are available for about $70 that use a computer USB port and generate decent clean audio.

Connecting samson co2

The Samson CO2 is a condenser (also called capacitor) mic, and thus requires phantom power to operate. In order to apply phantom power, you must have a board capable of handling an XLR connection, and supply phantom power. Usually there is a switch located near the XLR-input jack on your board - if you post your sound board type, more help can be given.
