The defrost drain line probably is located in the bottom rear, depending upon configuration. It is not a plug.

More info would be useful in helping you find this. A model#,age, ect. A drain can clog, or freeze. Some models are more likely to be one or the other. Sometimes you can simply defrost the fridge to solve the issue without having to take it apart while others might require a redesigned drain tube, probe or some other permanent repair.
Basically, they will be located behind the evaporator cover directly below the evaporator. This is very sharp and will cut you very easily! So caution must be taken when working around this area

You get to it from the freezer. Remove the back wall in the freezer and you will see it there. Unclog it with some wire or check to see if it has ice over it.

SOURCE: need a diagram of
I can't find a diagram but have a different model Amana. The drain tube on mine was located just above the drip pan under the refrigerator. It was a soft plastic/rubber and had become clogged due to something sticky that stuck it together. Caused the freezer water to back up and after cleaning the tube, I had to defrost the refrigerator completely to melt the ice blocking the tube at th top (in the freezer). Sorry I can't help with more info.

Get you a can of Computer air at walmart and purchase you a cork that is smaller than the tube make sure the cork will insert at least half way into your drain tube. Now preparation the air tube on the air can has a stem that either inserts into the can or is built in. Preferably purchase the one that is built on to the can. make the air can stem a hole in the cork small enough where you have to force threw the center of the cork the air can stem making sure it goes all the way threw the cork and blow it once or twice to make sure during the insertion it did not get clogged. after all this insert in into your tube and with several blasts form the can MAKING SURE CAN IS UPRIGHT of air this should dislodge any debris in the line clearing the clogg repeat process till clear.
