The only way to stop this silly calibrating every 3 prints or so is to unplug the printer, remove all toner cartridges, remove the drum (keep it in a dark place) then wait a ful 30 minutes.
Yet another solution is to bin your samsung printer and go with something that works. Smartphones makers are no printer makers. I and my wallet learnt that the hard way. Three Samsung printers, three went to the bin after 3~4 months...

The factory default settings for the S600 is to auto calibrate every time the printhead is closed and every time the power is cycled. If the media sensor is not positioned correctly for the media you are using the label size will not be detected during a calibration. You mentioned the media has no markings, doe sthe media have a space between the labels? If so, the media sensor may not see the gap in the labels adn therefor is not calibrating properly or not calibrating at all. Check the media sensor position. I have found that the media sensor must be at least 1" from the inner wall of the printer so it doesn't see the edge of the backing in oder to read the size of the labels.

No guarantee this will fix your problem, but the last time I experienced this problem it ended up being one of the toner cartridges. I replace her refurb. toners to OEM brand.

Sounds like it is set to do a label length check at power on. Go into your printer preferences and set it to "do nothing" on power up.

The fact that there appears to be no actual jam tells me that it is not picking up paper out of the tray. Try some alcohol on the pickup rollers, rubbing well. If that helps, replace these rollers.
