Use a good anti-virus software. There are many free ones, like the very popular AVG Anti-Virus Free Edition. Also, the Avast! Home Edition and Windows Defender are great. You can also try the Microsoft Windows Maliculous Software Removal Tool.

Download.com and the last one is a special program available only at the
Microsoft Download Center. It has to be downloaded, used and then removed from the PC. It is updated every month, so you need to make sure that you use it every month after downloading from the site.
To prevent damage, later on you must have backups of the files on your PC, either on hard disks or DVDs. You can use a back-up software like the preloaded Windows Backup and Restore Center available in Windows Vista. You can also copy your important software and data manually onto these media.
Don't backup now, or you are going to backup the virus too (if it is really there) and waste your time. Remember, the virus cannot do anything while the computer is switched off.
