Depending on operating system go to start then programs then accessories then entertainment then finally volume control. do you see microphone listed on volume control and has it been muted if so uncheck mute.

you can go to testmycam.com to verify it's working both the webcam and mic or you can install any messenger software (skype, yahoo) and connect your webcam and mic there as well
On Skype run the Audio / Video setup and make sure your webcams mic is selected as the default Mic.

You may just need to manually select your mic. Go to this free video mail site. Click on the mic icon and then try the choices in the drop down list. Record and play back brief recordings until you can hear yourself. Once you know the mic that works select it in Skype or other software program. It is usually under Settings, Audio.
