When 1 cylinder is misfiring, most of the time it is just a bad spark plug wire. Replace your spark plug wires and the problem should be fixed. If not, replace your spark plugs, but usually it is just the wires that need replacing.

Do a complete tune up.. Spark plugs, wires, distributor cap, and rotor button... If it has coil packs that sit on top of the spark plugs.. Replace the spark plugs and cylinder number one coil

ignititon coil is probably arking ,replace coil if bad.

SOURCE: misfire on number 5 cylender

Youm either have a fouled sparkplug [ should be changed @ 100,000miles ], a bad igntion coil on that cylinder or a faulty fuel injector. Most likely = a bad coil.

You might want to look for a vacuum leak on the intake manifold. You also might need to hook up a scope and measure injector duty cycle and compare it with a good cylinder. Also check wiring for chaffing going to the injectors and coil pack. I can provide you with pin out on ECM if you need.
