If one of the lifters are not working it sounds like you have a flat lobe on the cam shaft, you should be able to remove the valve cover of the suspected side and start the motor. Observe whether the valve rocker is moving the same as the others. If not, shut the engine off and check the bolt that hold the rocker to sure it hasn't stripped out or pulled out of the head, then take the rocker off and check for excessive wear around the point where the rocker is bolted to the head. If all of that checks out then the cam shaft is probably the culprit and will have to be replaced. You might visually inspect the suspected lifter the be sure it hasn't collapsed.
Replacing the cam shaft can be a major repair especially if you have never done that, it would be advised to have a shop do this repair unless you are bold and adventurous.
