I have a 2000 Lexus RX 300. The key can open and lock all 4 doors except the driver door. All connections seem to be OK

Get underneath the engine and attach a battery charger positive to the positive lead on the starter and a clean bolt on the engine or bodywork. In 15 minutes, you will be able to open the door.

Alternatively there is the easy way, use the emergency door key that is hidden in the fob.

I have read this 6 times and still not sure what the issue is? I have a 2005 ES330. To open all the doors with the key fob, you have to double press the Unlock Button. Pressing the Unlock button once, only opens the driver door.

Assuming you are not sitting on the key fob when this is happening, I think you must have a short in the wiring in one of the doors. Open the doors and check the wires that run from the body to the doors for any broken wires.

then you have a defective driver door lock actuator motor, to change this u must remove the door panel, drill out the rivet and remove the old motor and pop rivet the new one in place, this requires a special jumbo size river gun and a rivet head cutting tool.

The most likely cause is a faulty lock actuator assembly ( or a loose connection to it ) in the door itself. The inner door panel will most likely have to be removed, the lock actuator (or motor) tested and, if defective, replaced.

no. remove the disc from the nav disc unit and reinstall. you must have the key on or the vehicle running during the process

have you door lock module and the the brain in the key one of those is your problem have them check

fault in drivers lock probably micro switch or solenoid get tested

This is caused by the window express down module, if you quickly push the window switch down a few times does this correct the issue? If not have the express down module replaced.
