My blazer has two main misfires cylinders 4 and 5
No other History, symptoms, clues, things checked?
Would check the compression on all the cylinders; suspect 4 and 5 could be low if adjacent, and look for plug fowling (and whether oily, caked, or different from other plugs--all are clues).
Adjacent cylinders are suspect for head gasket between them (where it is narrow), look for white smoke (coolant) as well, and bubbles in radiator or even oil slick in coolant.
...would assume you already checked the spark uniformity
