The remote will lock all doors including the driver's side door. However, the driver's side door will immediately unlock and will only remain locked when done manually.

The body control module is responsible for controlling various electrical functions in a vehicle, including door locks. Removing the 50-amp fuse could have caused the module to reset, which may have resulted in the driver side door becoming locked.
To resolve the issue, try the following steps:
Turn the vehicle off and remove the key from the ignition.
Close all doors, including the driver side door.
Lock the vehicle using the key or remote.
Wait for a few seconds, then unlock the vehicle using the key or remote.
Check if the driver side door is unlocked. If not, try using the key or remote to lock and unlock the door again.
If the above steps do not work, there may be a problem with the body control module or the door lock actuator. In this case, it is recommended to consult a certified mechanic for further assistance.
1.  Close and lock all doors with the driver’s side power lock/unlock switch.
2. Â Insert key into ignition and remove it from the ignition key cylinder at least six times within 10 seconds. Your Hazard Lamps will flash if you have performed this step successfully. NOTE: Withdraw key completely from ignition cylinder each time. If this procedure is performed too fast, system will not enter programming mode.
3. Â Insert key into the ignition cylinder and turn to the ACC position.
4. Â Within 5 seconds, push ANY button on the remote transmitter. Your Hazard Lamps should flash. NOTE: Do not press the button more than one time in the above step. If the button is pressed more than one time, the programming procedure will not be successful.
5.  If there are any remaining transmitters (including the old ones), unlock then lock all doors using the driver’s side power lock/unlock switch and within 5 seconds, push ANY button on the next remote. Your Hazard Lamps should flash. Repeat this step for each transmitter (including any existing transmitters).
6.  Turn the key to the OFF position, remove keys from the ignition, unlock doors using the driver’s side power lock/unlock switch and open the driver side door.

It may be that water/moisture has gotten in behind the door panels...do you notice that it worsens in damp(er) weather? The car unlocks by itself if you leave the key in the ignition and you try and lock the doors.
Or somebody has the same coded fob (unlikely).
You might want to reprogram the remotes if you determine that the area behind the door panels is dry and there are no corroded wires, etc.
2004 Nissan Quest Remote Programming
Note: You must have all remotes on hand when performing this procedure. Any remotes not used during this procedure will become inactive.
1. Close and lock all doors with the driver's side power lock switch located on drivers door panel.
2. Insert key into ignition (DON'T TURN) and remove it from the ignition key cylinder at least 6 times within 10 seconds (this will need to be done at approximately one second intervals). You may need to insert and remove the key up to 10 or 12 times until your hazard lamps flash. PLEASE NOTE: Withdraw key completely from ignition cylinder each time. If this procedure is performed too fast or too slow the system will not enter programming mode.
3. Insert key into ignition and turn to the ACC position.
4. Within 5 seconds, push the LOCK button on the keyless remote. Your hazard lamps will flash. PLEASE NOTE: Do not press the button on the remote more than one time or the programming will not be successful.
5. If there are any remaining remotes (including working ones), Unlock then Lock all doors using the switch on drivers door panel. Within 5 seconds of locking and unlocking doors press LOCK button on the next remote. Your hazard lamps will flash. Repeat this step for each remote (including any working remotes).
6. When all remotes have been programmed turn the key to the OFF position, remove keys from the ignition, unlock doors using the driver's side power lock/unlock switch and open the driver side door. Test all remotes.
1) Key turns but dose not unlock door. This means that the rod that goes to the back of the lock has came off. This also means that is the reason door is not unlocking with the remote as it pops the lock rod inside the door which pushes the lock open. 2) Key does not turn lock at all but works passengers side door. This means that the locks has been changed in the door, but that doesn't sound like the problem as you say the remote doesn't open the drivers side door so more than likely the rod has came off the back of the lock
it is not the actuators, try a different remote, you may have an issue with the car security system/body computer not functioning on the unlock driver door command, one press of remote is unlock driver door and 2 presses is unlock the other doors.
this is a problem that can appear after your car battery died, cause the car didn't reset propely, so taking the battery out and putting it back in or letting your car die again would do the trick. I tried a lot of reprogramming procedured for my remote key, none of them worked, what did the trick though was simply unlocking the doors via the all door unlock button on the passenger side door, so simple.
