Where do i tighten my lifter up up at on my 2001 Kawasaki 300 Kawasaki 300 Prairie 4x4

Hello, What lifter are you talking about?... your engine does not have "conventional" lifters.. the valves are adjusted at the rocker arms/valves stem. If the engine is "ticking loud" . or not returning to proper idle quickly.. its time for a valve adjustment,,,> By a QUALIFIED professional!!!!!!

Why won't it idle without choke?

Fuel supply problem somewhere. How is the float adjustment and the float valve? I'd guess the level adjustment is too low.

Econ Quad Viper 70 4 stroke won't stay running unless choke is one quarter or more closed. Occasionally pops through carb and bogs down if accelerated.

Sounds like you need a valve adjustment, if it is popping thru the carb, the intake valve is open at the wrong time, this can be caused by three things, too much gas, timing is wrong (stretched chain) or valves too tight not allowing valve to close. I would adjust valves first.

Valve clearance adjustment

bluetraxx.com might have a service manual posted with that info. yamaha also has owners manuals free for download but not sure if they'd cover that.
