Your computer can only add printers it sees, whether connected or networked.
You didn't indicate if other computers can print to that printer. If your new computer is the only one, I'd suggest to reinstall your WiFi printer and try to add the printer again.

What brand and model of printer ?
You must have received my earlier message a second time. Since another computer is printing fine to the printer, no need to reinstall the printer. After you downloaded and installed the driver from the Canon site, did you see the printer in the printer list ?
Just reading about your printer. According to Canon, your printer is "networkable via an Ethernet connection, and offers an optional BU-30 Bluetooth interface, so you can print wirelessly from a PC or PDA". Do you have a wireless network, but the printer is connected via an Ethernet cable to your router ? Trying to clarify your setup.
Perhaps you have the wrong driver installed. You were uncertain if you have Vista x64. To check, click "Computer" from the start menu, then click "System Properties" at the top of the folder. After that, scroll down until you see "System type" and it should tell you 32-bit or 64-bit.
Yes ... try reinstalling your driver. But, remove the printer first if listed. Then, restart the printer and your Vista computer.

Be cetain to remove the printer icon (delete) before reinstalling the driver. To reset/restart the printer. Power off, disconnect the A/C adapter from the power source (wall, power surge), wait 1 minute, connect to power source, then power on, again.
Since you are getting a "new port" message, if you are trying to connect to WSD and are getting that message, this could be helpful: Quit all applications currently running. Click [Start], and then click [Network]. The [Network] window appears, and the device search begins automatically. Using as the NetWare Print Server / Remote Printer If the [User Account Control] dialog box appears, click [Continue]. The [Found New Hardware] dialog box appears. Click [Locate and install driver software (Recommended)]. If the [User Account Control] dialog box appears, click [Continue]. The [Found New Hardware] window appears. Click [Don't search online]. Click [Browse my computer for driver software (advanced)]. Next step Click [Browse...], and then specify a location for the printer driver. Click [Next]. If the [Windows can't verify the publisher of this driver software] message appears, click [Install this driver software anyway]. Click [Close]. If installation is successful, the icon of the printer connected to the "WSD" port is added to the [Printers] window.
Glad it worked. 1. WSD is the right port. com1-4 are direct serial connections to your computer 2. "should I delete all but the one printer icon that is now working?" If you have multiple PRINTER icons, delete the others. But, if the other icons represent OTHER functions, leave them. 3. "on the port tab, should I check the box that says "enable printer pooling"?" Leave the box unchecked.

Few things, in the add a printer, you should select Networked printer, not a local printer.
For the best results you might want to run the original setup disc and you may have to connect the printer to the computer with USB first, to re do the setup, the printer should continue to work normally with the older pc that its set up with.

If no printers show up in the list in step 5, then make sure the printer's wifi connectivity is enabled & configured with the correct information.

I am running windows vista home premium, and I have service pack 2.

Some additional information.
I am trying to add a new printer via wifi, not USB.
When I invoke the add-a-printer routine, no options for printers come up; i.e., not a single printer is listed. This is unnerving - I would like to have loads and loads of printers listed..
I do not know what to specify as a source for my printer.
I followed the procedure you advised. When I checked "printer I want isn't listed" I was shown a blank screen but there was a browse button, which I pressed. Nothing showed up.
I think your approach is not working. Any other ideas?
The other expert only passed me canned procedures. He did not respond to the primary issue: in Add Printer, no printers are shown, the list is empty! I complained about this after his Monday 1PM note, and I haven't heard from him in 2 days.
I would feel most secure if I could be helped to populate the printer list, including the Canon MX850, the printer I wish to add. Surely I will add other printers in the future. But, if it is easier to just add the one printer, so be it. Let's just get it done. Thanks!
An old computer can print wirelessly to the printer.
It is my new Acer Vista computer that cannot print.
While waiting, I tried the following: I went to the Canon site, downloaded the driver for this vista home premium system, 64-bit (is that right?), installed it, then tried to print a word file and got the error message "printer not responding". Does this help?
Sorry, your comment doesn't make sense to me, as it repeats the message you sent an hour ago that I responded to, saying an older computer can print wirelessly to the printer. Also, you suggest that I reinstall my wifi printer, but this is nonsensical if I have no printers listed in the add a printer list. Or do you mean find the startup disk for the printer and run those routines?
oops, disregard my last comment I didn't see your firewall statement. Did you get my notification that I downloaded the canon driver and got a printer not responding message? I'll turn off the firewall now and try.
I turned off firewall (webroot), went to control panel -> printers -> add a printer, and the printer list was not populated. The canon printer was listed with "error" by it.

No, I did not see the printer in the printer list, although it was listed when I did console -> printers but with "error" beside it.
The printer itself is connected by cable to the router. The old computer connects to that printer wirelessly.
it says 64-bit, so I had the right driver. Should I perhaps download it again, in case something got corruped in the download?
As I said, it's 64-bit. Just to be sure, I downloaded the 64-bit driver for the mx850 from a different site, installed it, it installed fine.Tried to print and got the "printer not responding" message. ...
Thanks very much for your help. There were 2 64-bit drivers one identified as a "network" driver, the other without a descriptor. The first was much larger. I had installed the second one before and this time I installed the first, the network one. This time, after installation, there was a msg "searching for printer" and it did find the mx850, I clicked on it to continue the installation and got an error message "new port could not be connected", and it offered "retry." So there is progress.
Oh, I didn't restart the printer. I don't actually know how to do that - turn it off then on again?

OK. I just printed something! But when I do start->printers, the window shows multiple copies of the mx850 some as fax some as printer (I guess these came from multiple installs, when there were errors and I would start over). I picked the one that said printer, checked its port, and it was set to "file", so I reset it to WSD, and that is when I was able to print.
Questions:
1. Is WSD the right port? any reason to pick com1-4?
2. should I delete all but the one printer icon that is now working?
3. on the port tab, should I check the box that says "enable printer pooling"?
