Check the BIOS settings. It is possible that USB is disabled hence it will not work. Prior to boot up to OS, check the screen and it will display what key to press (normally F1 or F2 of F10) so you can go to the BIOS settings. If your desktop is connected to your company network, it is possible that your IT has disabled the USB via the registry, in this case you cannot do anything unless you have administrator access. Another possible cause is a defective motherboard.
