The Dell Dimension E520 won't boot up but the amber lights come on...

Make certain all the connections to the motherboard are good. Sometimes they won't power up because the power supply isn't good enough for the job.
On some motherboards you have to clear the Real Time Clock (RTC) and clear the cmos. This often requires the removal of the battery and the reposition of a jumper connector.
You should consult the manual of the motherboard for more information.

When installing memory in this motherboard do you have to fill all 4 slots?

No, a minimum of 1 memory card is needed for the computer to clear POST and start up.
You can put as many as it can afford. For your computer the maximum memory is 4GB i.e. 4 cards of 1GB each.

I removed memory while computer was on, now it wont power on and boot a solid amber light stays on. how can i get my computer to work

While PC was on? You might have damaged the whole motherboard.

Try removing all non-essential devices and booting it to minimal. Try another stick of RAM. IF still nothing, the motherboard will need replacing,.

Power loss

have you folks removed/installed memory with the computer plugged in ... but not powered on ???
newer systems maintain standby power on the memory .. if you have .. you could have blown a DIMM ..
I'd remove all installed memory .. then power up and see if the systems behaviour changes .. if it does .. and your system supports it .. add a stick of memory at a time untill you recreate the problem ..

Well hate to say it but that sort of problem results from a faulty motherboard or processor so if you say that you have changed the motherboard then you problem lies with you processor chip.
