Yes, it will work. Obviously, you're not going to get any autofocus with the lens. Also, you will lose your exposure meter. You'll have to shoot in Manual (M) mode without any assistance from the camera's built-in light meter.

This lens, like all mirrors, has a fixed aperture. Your only controls over exposure are the shutter speed and ISO.

Will a opteka 500mm mirror lens fit onto a nikon d3100?

When you go to the Opteka page here http://opteka.com/500m.aspx
You can select "options" for the camera company (there are 6) which you have. It may mean that the lens uses an adapter for the different pin and thread patterns, so the lens if bought new will fit with the selected option, a used one, you may need to get an adapter.
Best bet is to try before you buy.

How to insert filter in 500mm mirror lens

Look inside the back end of the lens (the end which attaches to the camera). You'll see a threaded ring around the optical part. The filter simply screws onto the threaded ring.

Skylight lens

The small filters should screw into the back of the lens- also there needs to be one of them on the lens at all times (it's part of the lens design), so there may be a filter already there that needs to be removed before you can put any other in it's place

Opteka 500mm f/8 mirror lens for my DSLR and it's says no lens

You need to turn the mode dial to Manual (M). You won't be able to use the camera's built-in exposure meter at all with this lens. You can take an exposure reading with another lens to use as a starting point, then review the pictures and histograms to fine-tune the exposure.
