You have to plug it into the computer, look at the list of music on your mp3 player, highlight the song, and push the delete button. It may be a little different depending on what you use to manage your music (iTunes, Windows Media Player, Rhapsody, ect).

I have deleted the songs from the file on my computer, but my sansa e250 is unable to accept new songs in place of the ones I deleted
