Your problem cannot be classified as 'minor': you have probably been infected by a nasty virus.
In order to do this 'manual' delete of karlewsa.exe or nay program that is 'used by another process', you need to:
ps. The best way to kill any virus is to boot up from a CD containing an up to date antivirus/antispyware utility that has been created on a clean PC.

Hi, I've found that the virus spreads when you plug in any form of removable storage to the affected computer.
I've also found that the Virus can be easily removed from said removable storage when plugged into a Mac; as the hidden folder "OLJA" turns up, which contains the karlewsa executable.
I'm not sure how to remove it from the affected PC, but I hope this helps to prevent the spread of the virus.
