How do I make this happen on the calculator x^2-64=0

Yes you can enter basic or complicated expressions, you just can't enter equalities nor equations. The calculator cannot solve equations. When an expression (without an = sign) is entered, pressing the ENTER key will evaluate it.

You cannot. To evaluate an expression (not an equation or an equality) key it in and press ENTER.

There isn't one. You have to use the old eyeball. If the number on the screen is positive, leave it alone. If it's negative, change the sign.

If you mean matrix, then I am sorry, but neither the TI-30Xa nor the TI-30XS calculators handles matrices.
