Place a few drops of vinegar into the screw hole and wait a little bit. Try removing the handle again, sounds like it is just stuck on the cartridge stem. The handle should come off first, and then the bonnet. Here's the parts breakdown for your faucet from Kohler.

There's usually a plug button (on the top or around the base of the handle), remove that and there will be a small screw underneath.

I took the screw out of the handle on my kohler k 12265 4 widespread faucet but the lever will not come off. Is it stuck or do I need to do something else/ I don't want to break it by forcing it off.

It will come off with the right amount of force. If there was a screw in the top then that is what was holding it on. Just make sure there is no allen screw on the side which there should not be. First spray some penetrating oil on the screw hole and let it sit for an hour. Then take a large flat blade screwdriver or small pry bar and pry on the underside. Keep moving around the outside in a 6-12-3-9 pattern until it releases. Try not to damage the finish. Eventually it will come off. I assume it is leaking so you have no choice but to keep prying until it comes off.

Kohler Fairfax single control kitchen sink faucet

Remove the air-irater at the end of the faucet where the water comes out. That is where 99% of clog are on faucets. If that does not do it then you need to replace the cartridge.
