It's been such a long time since I last used my Janome Harmony (I'm not really feeling very harmonious) 2049 that I can't remember how to get the thread up to the needle. I know laugh all you want....but I've tried everything...help!
Your question suggests that you want to have the bottom thread ready to sew, and that you DO know how to thread the needle................so.............
Thread the needle...........pull an excess of about 75mm (3") to the rear of the machine and keep hold of it,...........manually turn the handwheel towards you - anti-clockwise - one revolution (finish on the needle up position)....clear the 2 threads from under the foot and you are done.
Of course this will only pick up the bobbin thread if the machine is correctly timed, if not, then you will need to look at re-timing the needle to the hook.........see how you get on with this first.
