When you plug in the USB, it should make a sound on your computer, I find that if I move my player, it can dissconnect quite easilly.
If you have windows, Media player will sync up and show it in the player

Suddenly the sansa will not synch with Windows Media Player.

When you attach your Connect via USB to your computer, verify that the Connect is displayed as a "portable media player" under "My Computer" in Windows Explorer.

If it isn't ... try a different USB port. The Connect requires a high-power USB port, which is typically found on the back panel of your computer.

If this doesn't work, try the Connect on another computer (just verify that it shows up under "My Computer", as I mentioned). If it does, you may have a problem with the installation of WMP on your computer ... try re-installing WMP, and make sure you install the latest version of WMP. If the Connect isn't recognized on the other computer, it's possible that there is a problem with the USB cable - get a replacement.

Is it still connected to your computer and mounted in iTunes? If so the ipod will show up under devices in the iTunes sources list. You need to click the eject icon before you can use your ipod while it is connected to the computer.

Here is one answer, from the apple website:

Syncing music manuallyIf your prefer not to automatically sync music content to your iPod, you can setup manual syncing.
  1. Connect your iPod to your computer and open iTunes.
  2. Select your iPod in the DEVICES section of the iTunes Source list.ht1351_7.jpg
  3. In the main iTunes browser window, select the Summary tab.
  4. Select the "Manually manage music and videos" option. This will deselect the automatic sync settings for the Music, Movies, and TV Shows tabs. Automatic syncing for Podcasts, Photos, Contacts, and Games can be individually disabled by selecting each of those tabs. Note: When manually managing iPod, disk use is enabled and iPod must always be manually ejected in iTunes before disconnecting.ht1351_8.jpg
  5. Click Apply.
  6. To manually add music, audiobooks, movies or TV shows to your iPod, choose one of these categories under LIBRARY in the Source list
  7. Select and drag items you want (to choose multiple items, Command-click items on a Mac or Control-click items in Windows), then drag them onto your iPod icon in the Source list.
  8. To add a playlist, select it in the Source list and drag it onto your iPod in the same ways as a song.ht1351_9.jpg
  9. When you are ready to disconnect your iPod, select it in the Source list and click the eject icon.

Does the device show connected? If so check the device manager for any red or yellow errors. If there are errors, right click on the device and select un-install. Disconnect the device from the computer, wait a few seconds and reconnect. The device should re-install the drivers and device should be recognized. Best of Luck.

Try changing the "mode" on the device. Settings>Mode or USB Mode>MTP or MSC.* *(if you have thr 'R' version, your options will be Rhapsody or Plays for Sure) If you transferred music in one mode, you are not able to see it in the other mode. Best of Luck.
