This could be either mechanical or software. Try the right mouse click...does it work? Try another external USB mouse. If a known working mouse doesn't work, your computer may have a virus preventing you from clicking.Else, your mouse has a broken or dirty switch.
Some mouses stop responding if you do not use it for a few minutes. This is usually a battery saving feature. Usually all you got to do is use the scroll button to reactivate the mouse by scrolling up or down. This will bring the mouse back to life. When you change the ports you are actually doing the same thing (reactivating). Also check whether battery needs changing.
if you plug usb keyboard onto your laptop it will probably turn the touch pad off.
If no other keyboard is attached there is usually a switch to turn off the touch pad.
also if you attach a mouse this may disable the touchpad so you dont move the mouse pointer when you are typing
