Dos won't be able to use most of the hardware.
I've talked to a few guys who still maintain DOS programs what they do is install a DOS emulator in thier windows based system.
Here is a link to a page that has 5 emulators that are free. You may find lots of others as well. I don't know what you are looking for ie joystick, sound, graphics etc so I didn't try to find anything specific for you. Since these are free and probably lots of others I would grab them and try them out until you find one that covers what you want to do. The X128 one runs under XP and Vista so it might be one to start with.
Using an emulator will let you use a normal windows setup and let you develop your DOS stuff on the same machine without trying to solve how to get dos to read hard drives etc.

The best way to do this is probably use windows and run dosbox for windows. What it does is it emulates DOS and lets you run any program that requires dos. Because your computer doesnt have a LPT port. and nowadays only drivers for dos or newer operating systems come with the card. installing the driver in windows then running dosbox, it will see the LPT port.

To run the check disk utility you will need a repair disc or
a operating system disc. Put the disc in the cd drive and boot from
it. You want to get to the Dos Command Prompt (Black screen with white
letters) and type the following command: chkdsk /r
Check disk will run well over an hour or two and you will notice the percentages going from high to low, this is normal. Let it run to completion. Then reboot machine.
If check disk fails to solve your problem you might have to reinstall windows. Try doing a Repair Install that way you don't loose your data, documents, music, pictures, videos or programs.
In windows xp you get to the dos command prompt by selecting R for repair when the options appear.
In Vista and Windows 7 you want to get to the recovery console and look down at the bottom of the window for the Dos Command.
