Well it all depends on the tower and motherboard. Typically not located on the motherboard itself, it is usually plugged into a port on the motherboard port and extends to the top of the tower or front of it. "Front panel USB Ports"

Front 4 usb ports and microphone and headphone jacks don't work but the rear usb and speaker jacks do work. I've had the computer new since 2009 and they have never worked.

it sounds to me like the wires from the front panel connectors are not connected to the motherboard inside the computer case, open the case up find the front connectors wires inside and find out if there are any connectors on the motherboard to plug them into.
the reason the rear ones work is because they are soldered directly to the motherboard.

    The lighter gray, curved front panel of the computer can be flipped up to expose two usb ports and a headphone jack. Plug your headphones into the round hole.

    This dell computer does not have a built in microphone.

    It has a microphone input jack on the right hand side of the base just to the rearr of the headphone jack and just in front of the 2 USB ports.

    Please clarify, Where is the Headphones connected? Front or back? if you tried it on the Front port without success, Plug in the speaker jack to the port where you did connect the Headphone and verify it's working or not. You can also try the headphone on the port where the speaker is connected.

    Front panel connectors/jacks is not working

    OK--usually what you will find from the front is 3 connectors.
    One will be a solid block that has one blank hole which is the key. That will plug into the corresponding pins on the motherboard--usually toward the back of the case.
    There will be another other wire that terminates in two plastic connectors. These are the USB connectors. Sometimes the connectors are in two parts. One is a 3-hole and one is a 1-hole. Usually you will find two sets as they sort of go together.
    The pin configuration on the motherboard looks like this ::::.
    The single pin on the end is not used. The other ones are connected with the plastic connector on the first three starting at the left, with the end marked VCC to the left. The fourth pin is the ground which is usually the separate connector.
    The second USB connectors are plugged in right next to the first--so each block of pins controls 2 USB ports.

    Headphones with mic / external speakers

    Does your computer have front audio ports? If so, you can just plug it in there. Otherwise you need a single male 2 dual female converter to plug into the speaker port.

    Answer me this. You're case is an Antec tower? The motherboard is the Asus P4P800 SE ? Is this correct?
    If you have you're manual you can see what goes where on the board for front panel connector page. 2-31. (This diagram shows you how to connect the front mic & speaker).
    To connect the Antec's front USB ports plug the case USB front connectors onto USB56 & USB78 shown on page. 2-30. Most case wires are already set within their USB connectors & the Antec's front USB connector should fit right on the mother board's pin's.     Headphone jack not working.

    If you have a headphone jack at the front and the back, it may be a wire to the front panel is not connected properly on the Motherboard.
