You don't say what type of boiler you have, but as a guide...

Three port valve may not be operating.

Heating Pump may not be operating.

Room thermostat incorrectly set or not working.

Start with the room stat as it controls the valve and the pump..

More information about your system is needed to help you further..

Need to separate heating and hot water - just have hot water on

You need a heating engineer to come in and do this.

Why does it take two bowls of water before water gets hot on combi boiler

The water isn't maintained hot (unless the central heating is on) in a combi boiler. you will, therefore have to turn the water on, which activates the burner. The burner will then have to heat up the heat-exchanger which then supplies the hot water through cold pipes. Once the pipes are warm, you will receive hot water.
