Sounds like either a bad plug wire, or coil pack. This is about all that would cause the misfire.

First, check the fuel lines for cracks/breaks.
If they are o.k.,
then you will need to disassemble the carburetor again. Make sure the small screen in the carb. is free of debris. You should be able to see daylight through it. Use
a Den Tek Brush, (http://www.drugstore.com/products/prod.asp?pid=165357&catid=1152), to clean the small portals in carb. These portals can become blocked. Reassemble and start.
(NOTE: If the cleaning doesn't work, then buy a carb. kit and replace all appropriate parts.)

If the crankshaft/flywheel alignment pin is sheared the firing will occur at the wrong time. Check that by removing the flywheel.

the fuel could be good but still it wont spit check the carbs when you throttle or kick start if thats possible if that is good try some starting fluid it should start up and the fuel pump should get out the obstruction if theres more problems it is one of many things either bad fuel pump, your fuel line is disconnected, blown headgasket idk the psi specs for the 250 but that sounds good i know for the 550s its around 120 and so on

Will it start if you pull the plug and spray starting fluid in the chamber? if so then I'll bet you have a blown diaphragm in the carb or a plugged fuel filter or a bad gas line. hope this helps. Jeepfxr
