My Weber Spirit grill won't light. It will light manually, so I know it is getting gas, and the igniter sparks so I don't understand what the problem can be.

Since there is some dismantling of the grill required, to remove the burners. I have included a link to the Owner's Manual for you grill.You can download and print it for future reference.
Bill, I suspect that the third burner needs to be removed and thoroughly cleaned. As does the venturi valve on the open end of the burner. If however, the 3rd burner is the one that ignites from the cross over tube burner, you will have to clean and/or replace the cross over tube burner.
To assist you further, I have included a link to the Owner's Manual for your model grill, Just in case you have misplaced yours. You can download and print it for future reference.
Do you mean it won't match light or the ignitor won't spark and light the burners? Or both?
If it won't match light or the ignitor sparks, but the burners don't light, that means you're not getting gas to the burners.
Check to the tank valve and make sure it's turned ON. Also, make sure that there's gas in the tank. If both of those check out as OK, turn the gas tank valve OFF. Disconnect the hose from the tank and wait 5 minutes. Reconnect the hose to the tank and turn the tank valve ON VERY, VERY SLOWLY (Slow motion, slow). Once the valve is fully open, light the burner farthest from the tank first and turn it to HIGH. Did the burner light and the flame height looks correct? If so, light the remaining burners following the same procedure. Now, are they all lit and the flames heights look correct? If so, you solve the problem. Just remember to always open the tank valve very, very slowly.
if the burners failed to light. Remove and clean all the burners, as well as the venturi valve on the open of each burner. Spiders like to build nest in the venturi valves.
if cleaning the burners did not solve the problem, you will need to replace the grill's hose/regulator assembly. You can find it in any home center or hardware store that sells gas grills and accessories, for about $30.00. Take you old one with you for comparison purposes.

Is the igniter not sparking or is gas not getting to that burner? Have you tried to match light that burner? Those three things is where you start troubleshooting the problem
If the igniter is not sparking, it could be that the igniter is covered with cooking grease or the wire to the igniter has come loose either at the igniter or the igniter module. And simply needs to be reattached (pushed back on).
If the burner won't match light, than it's not getting gas and should be removed and thoroughly cleaned. And while you're at it, remove and clean all the burners and the venturis valves.
Removing the buner burners on a Ducane grill is a bit of a task, so I have included a link to your Owners Manual to assist you. Pay particular attention to the section Annual Maintenance, starting on page # 28.
Unfortunately, there's nothing you can do. According to Weber, there are no conversion kits, as your grill is not convertible to propane. Like wise, a Weber propane grill is not convertible to natural gas.

Weber sells conversion kits. Basically, you need a new regulator and manifold to accommodate LP instead of the existing regulator/manifold setup for natural gas. Unfortunately, Weber does not sell their parts online. Their parts phone # is: 877-731-8081
The area where the igniter strikes the spark probably is covered in grease and needs to be cleaned. Also check battery, and make sure wire is connected and getting good contact.
If there is no click, it is most likely the igniter module is bad. The igniter module should be available at any Home Depot or Lowe's; in any case, your owner's manual will show you how to remove and replace the igniter module.
