What Make and Model?
One fix would be to jury-rig a spool pin using a wood dowel or metal rod (metal coat hanger piece.) Put it behind where the broken pin is and duct tape it in place. Would look odd but would work.

How to remove broken horizontal spool pin of Singer Sewing machine model 6550 and replace with new spool pin?

It appears to be just "hooked" on according to the photo, and you can get the replacement part for $4.00 using this link: http://www.sewingpartsonline.com/spool-pin-singer-3709-sewing-machine.aspx.

However, before you go to all that trouble, you can also just use the auxiliary spool pin used for twin needle sewing which came with your machine but can be purchased from the same site for about the same price.

I don't use either -- I just put the spool in a cup behind the machine and thread from there. See? One way or another, you're back in business!

Take the machine into a repair shop. You won't have all the tools or the parts to make the repair properly.

Janome Rx 18S just purchased one second hand online and 1 of the spool pins are broken off. What was the purpose of the 2nd one and where can I find a replacement?

they are only used if your sewing with double needles to hold the
other spool of thread,, most people just buy a thread holder to set
at the back of their sewing machine
here's a link for one for the machine, but a generic spool or cone
holder is just as good,

Do not know how to replace spool pin and spool cap

It looks like it might just snap into place. Of course that means you have to remove the broken bits which may require a drill, small bit and needle nose pliers.

Broken plastic part within which thread spool pin is inserted

You can search for:

Make Model parts diagram

to see what replacement parts are available.

If not, you can probably devise something to improvise. I've used plastic drinking straws as spool pins, or plastic chop sticks.
