SOURCE: Zebra S600 printer continues calibrating
The factory default settings for the S600 is to auto calibrate every time the printhead is closed and every time the power is cycled. If the media sensor is not positioned correctly for the media you are using the label size will not be detected during a calibration. You mentioned the media has no markings, doe sthe media have a space between the labels? If so, the media sensor may not see the gap in the labels adn therefor is not calibrating properly or not calibrating at all. Check the media sensor position. I have found that the media sensor must be at least 1" from the inner wall of the printer so it doesn't see the edge of the backing in oder to read the size of the labels.

If this unit has the LCD screen you need to Press Setup. Then Press Next till you get to Media Power Up and set it to FEED. Next go to Head Close and set it to FEED also and then save settings. It will only shoot one label out instead of Calibrate. Which is needed when they install new lables if its the same type of label all the time really don't need to Calibrate every time you stick new labels in it.

some labels are hard to calibrate, nee to calibrate the sensors with manual calibration.
1} go to the MEDIA AND RIBBON CALIBRATION,
2}take 2 or 3 labels from the base paber, load the way to the based paper stay unther the sensors, 3}close printhead.
4} press ok.
5} Move the ribbon out from sensors and close the printhead.
6) press ok
7) wen printer ask RELOAD ALL, load the labels correcly and the ribbon, close the printhead and the top door, and press ok. The printer run the calibration it self.
wen finish, saved (press setub twice) and test.

until you get to "MEDIA POWERUP" Hit SELECT Use the +/- keys to enter the
password (1234) Hit SELECT - you should see a
small - and + button on the screen on
either side of NO MOTION (if not, repeat previous steps) Hit the + sign until you get to
CALIBRATION Hit SELECT to exit MEDIA
POWERUP Hit the + button to go to the
next item HEAD CLOSE Hit SELECT - you should see a
small - and + button on the screen on
either side of NO MOTION (if not, repeat
previous steps) Hit the + sign until you get to
CALIBRATION Hit SET/UP EXIT (twice) to save
changes (permanently) Open and close the printhead to
check to see if it worked!!!

I assume you have the transmissive sensor positioned so that it finds the label edge for both label types. i.e.the sensor is not looking at the vertical gap.
I also assume that the backing paper for both label types, if the liners are different then the sensor may be struglling to get adequate light through varing papers.
I would try to do a test print from the printer control panel on both label types. this will identify if it is indeed a sensor problem. If it work well through printer text, then the only remaining thing to look at would be the software/driver settings. Make sure the label length, sensor-type is correct for both label formats, and in the driver.
