My laptop will not pick up my  portable USB speakers.  Is there a sequence to making them work?  Do I first plug them in, turn on computer?  Which USB should I use the ones in the back or on the side?

may be u r laptop is not capable to provide right voltage to u r speakers............
try another source of power supply man........
dont forget to tell me u r problem solved or not
