A "USB" cannot be formatted. A USB is simply a port, one of several methods of connecting a device to a computer. There is also serial ports, parallel ports, fire wire ports, infrared and blue tooth. There may be other ports, the ones I mention are common.
If you have a device you have connected to your computer through a USB port, Windows newer than 98 should detect the device. If the device requires external power, make sure you are supplying it. (Did it come with a power cord or a transformer and power cord?) Find the device. Click on My Computer and see if it is listed. You have given scant little description of the device so I am slugging forward in the dark. If this is a mass storage device or a memory stick or a thumb drive and is a recent device (new), it should be formatted already and should open when you click on it. If it is an older device (an old Cruizer for example) you can go to the Cruizer web site and get instructions on how to remove that application and use your stick as a normal memory stick. If it is neither, right click on the device, one of the options should be "format" ... select format.

Normally you can format a drive by seeing the drive in "My Computer", then right clicking on the drive itself. A window opens up and a choice about hal way down the menu is "format" Be careful that the selected drive is what you want to format, as once formatted, all data becomes lost.
In order for you to format the pen drive you need to connect it to your computer then once the computer detected it and a auto play window came up don't open it, just go open my computer and check if the pen derive is there if its showing right click the drive and click format and once its done the drive is now clean of virus.
You will probably need to remove the drive and use another computer to format it.
You can buy an adapter to connect it to another pc as a USB drive and format it this way.
They are not very expensive.

You can not format the Windows Drive (referred to as windows partition) while the windows is running.
CAUTION: FORMATTING A PARTITION OR HARD DRIVE WILL RESULT IN ALL THE DATA BEING LOST.
But you can format any other drive or partition, to format a drive or partition other than the windows', open My computer right click on the drive icon and click format, press all the OKs and your drive will be formatted in a while.
To format whole hard drive, even the windows partition,

Now back to business, to do a complete format you need a bootable cd, If you have your recovery disk or even a bootalble windows disk you can format everything.
Lets assume that you have a windows bootable cd, insert the disk while the laptop is starting (make sure that the computer boots up with the cd), let the setup load its files, then follow the instruction until you reach to the point when you can see your hard drives and its asking for formatting press F to format the drive which is highlighted.
