Check that the USB and LAN functions are enabled in the board's BIOS setting. If everything is set properly and they still don't work, you could have a bad motherboard. Check into the warranty if the board is new enough, or just use expansion cards to replace the lost functions.

USB ports won't work on my ASUS PA 246, any idea why?

Which ports? front or back. The back ones could be because you don't have the drivers installed. The front ones could be because they aren't connected to the motherboard. If none of them work then it is likely to be because your USB controller has failed. You can fit a PCI card to provide some if necessary.

Run "Windows Update", to see if it will find updated device-drivers for the specific USB hardware on the motherboard.

Access the ASUS web-site, and search for 'P6T', and search for USB device-drivers for Windows 7 -- if none are found, try the device-drivers for Windows Vista.

If the usb ports don't seam to have power, I suspect that at some time something was plugged into one of the ports, either at an angle or upside down, shorting the +5 vdc supply. Usually the supply voltage is fed to the ports thru a "zero ohm resistor", basically a fuse that blows protecting the power supply. You may be able to find and replace it but much easier is to install a 4 port usb pci card. They're cheap and you can go to 2.0.

How to verify your Network card is working properly

If you are unable to resolve the problem with your intergrated lancard then you can add additional LAN CARD on your computer or you can also buy USB to LAN CARD adapter.
