Check the power supply. Re-install the drivers if applicable. Can you hear the hard drive spinning, does a light come on? Also try a different cable from the device to the computer. Go into Device Manager and see if their is some info there. Check the bus that it uses (USB, Firewire, etc) in Device Manager. If you know how, remove the hard drive from the case and install it on a bus inside the computer case. If it works this way you know the problem is with the external case or power supply.

320GB Hi-Speed USB 2.0 Silver Edition Desktop Hard Drive

Some usb drives - i.e pen drives - bring up the same message when they are inserted into a usb port. Windows usually installs the necessary software from within your computer. Other pen-drives are plug and play. Much depends on the operating system you have and the age of the usb pen.
It's probably just a question of getting updated drivers for your desktop hard drive. Right-click on the 'my computer' icon. Choose 'properties' Then choose 'device manager' or 'hardware'. Locate where your desktop hard drive is listed and open it. Then choose 'update driver'. Your computer should then search itself for updated drivers (which it won't find..) but it should also ask you if it can access the internet to search for drivers.
Alternatively, go to a website where you can download drivers yourself. Try http://www.iomega.com/support/documents/10425.html
Be aware that your desktop hard drive is now quite old. It won't last forever .. If you've got important stuff on it consider buying a new desktop hard drive and transferring everything to the new one.

Externe hardrive not working

Check the cable and power supply to the external drive. Has it been format for your OS? What type of interface, i.e. USB, Firewire, SATA, etc.
Goto Control Panel > System > Hardware > Device Manager.
See if the interface for the external drive is shown and working.
Open "Computer Management Console" and select "Storage". Do you see the external drive?

Hi actually i connected twice my External Samsung SP1644N to my laptop but later i cant find it even in my device manager. while i receive Cypress AT2LP RC58 with yellow alarm under other device! what...

Cypress AT2LP RC58 is the logic controller installed on the control board of the hard disk - if you get this label in Device Manager it means that the control board has failed and the firmware of the disk is gone = the disk is dead.
Junk it and get a new one.

Why can't I access anything that was copied onto my Toshiba 500G external harddrive. The computer doesn't recognize the program that made any of it, even though it's the same computer.

It is very possible the voltage on your computer's USB port is not handling the load. I would buy a "powered" USB 2.0 hub, plug it into your computer's usb port and plug the harddrive into the powered hub.

Having a similar problem with the La Cie 500gb. Powerbook G4.

The problem that has been inherent with all LaCie external harddrives, is the interface in the case itself.

Interface:
All external harddrives have a Style of harddrive that is used in a laptop, or one that is used in a desktop computer.
Laptop harddrives are 2-1/2 inches across in width.
Desktops are 3-1/2 inches across.
Your LaCie will have a 3.5, (3-1/2) inch desktop style harddrive, inside the case.

There are two main Types of harddrives.
IDE and SATA. (There is also a newcomer. SSD. SSD isn't being used in LaCie external harddrives at the moment, to my knowledge)

Your LaCie will use a SATA harddrive inside it.
This is an example of an average SATA harddrive,
http://en.wikipedia.org/wiki/File:Hdd.jpg
The photo of this harddrive is tilted at an angle, that doesn't readily show the interface connection, but it helps to explain.

At the bottom of this harddrive in the photo, and to the left, is the interface area.

Viewing the harddrive, look at the four metal pins on the extreme right of the harddrive. To the left of these pins is the Jumper pin area. (Has a white jumper on two of those pins)

To the left of the Jumper area is an opening, and to the left of that, is a long slot. The first small slot area is for the 7-pin Interface cable.
(Data cable) The next slot to the left, is for the 15-pin power cable.

This 7-pin Interface mates up to socket holes in a connector, of the case of the LaCie external harddrive.

It is this interface connector, inside the LaCie external harddrive case, that is the problem.
Solution?
Buy a quality, in-expensive External Enclosure, take the harddrive out of the LaCie, and install it in this enclosure.

Examples of 3.5 Sata External Enclosures,
http://www.tigerdirect.com/applications/category/category_slc.asp?CatId=2780&name=3.5-eSATA-enclosure&Nav=|c:2778|&Sort=0&Recs=10
(From $15 to $35, is a good range for a decent external enclosure)
