Error message: The folder "iTunes" cannot be found or created, and is required. The default location for this folder is inside the "My Music" folder.
I have two hard drives and recently a service man moved the iTunes to the alternate drive because it was saturating the 'C' drive.

Go to "Preferences" in iTunes and change the music folder location to the location on your new disk drive. Click the boxes as shown below:
You already tried removing and reinstalling iTunes (the program, not the music), right?
