I had the same problem with My Espon Perfection V500. I called tech support and they had me do the following sequence.
1. Turn off the unit
2. Unplug the lid from the base
3. Replug in the lid.
4. Turn the unit back on
5. Go into the Epson software in Professional mode, Configuration, Other and select Reset All
6. Rescan the picture.
It worked after that. Of course it also lost all my preset sizes in my Custom box and any other settings I had. I suspect the lid plug is what the problem was.
Another word to the wise. To test it I put in a picture that was laying on my desk and rescanned it and was disappointed to see the line in the result. A few minutes later I discovered that this particular line was in the original picture.

Hi! You're looking for an ac power adapter. I'm including a link to a website where you can purchase one. Just click on the link, or if it doesn't work, copy and paste it into your browser's address window. If this helps, please rate this solution. Thanks! Hal

Today I installed VueScan on my Win7 (7264) X64. After that the 1250 was still not recognized. Go to start and execute/run devmgmt.msc. You will get the hardware list with a yellow marker on your scanner driver. Double klick on that icon. Now you need to go to the second tab (driver program if I translate it) and second button (renew if I translate it) where you can install drivers for the device. Tell windows to search on your computer and point to the VueScan directory (directly on C: , not in programs). Now Windows 7 (7264) X64 will find the driver and your Epson 1250 is recognized. :-) Tested and working.
This same method worked for installing the AlexP drivers for my PS3 Eyetoy on windows 7 (7264) X64. Also had to point manually to the AlexP dir and then the drivers dir.
I assume this method wil also work for Vista X64. Haven't tested this, I only have Vista X86.
Sorry for my bad english btw. Not my original language.

Disconnect and remove the lid. Remove the two screws on the back of the base. Then use a screwdriver or flat blade to slightly open the back seam. Beginning at the back, run a blade up inside the upper part of the side seam. You will need to release three catches on each side. Once the sides are loose, raise the back of the top, while pushing forward, and the two snap catches at the front will release. The top then lifts off and you can clean the glass. Reassemble in reverse order.
