Only do this if your iPhone is stuck at the Apple logo or you've had some major problems. If possible do a backup of your iPhone first by right-clicking it in the left sidebar in iTunes and clicking backup. It may take a while.
First turn off your phone, either by holding the power button until 'Slide to Power Off' pops up or if you're phone isn't booting, keep holding the Home & Lock buttons until the phone is completely off.
Now hold the home button while plugging your iPhone in to your computer (and keep holding it). iTunes should pop up saying that it's detected an iPhone in recovery mode. Press OK and then press Restore. As long as you've synced and backed up recently, all of your SMS, emails, and contacts should still be on the phone.

Connect your phone to the computer and open iTunes. Hold both the home key and the power button until the iPhone turns off. Continue to hold them both for 5 more seconds. Release the power button but continue to hold the home button for 10 - 15 more seconds. I tunes should detect the phone in DFU mode. Restore firmware from this mode.
