From the sound of your question you are trying to move your whole system on to a new drive.
It is not possible to do it using a simple copy. There are many reasons for this, but the main one will be that files are open and cannot be copied.
The best thing to do is run NTbackup.exe and use it to make a backup of the disk. Alternatively you could use a product such as Drive Ssnapshot (http://www.drivesnapshot.de/en/down.htm) to take an image of the disk.
