Two possibilities are the port configuration and the type of serial cable.
The printer has a feature that will try to match itself to the PC's configuration. Press and hold the feed button until the green status led flashes three times. While the led flashes send a ZPL format to the printer (hopefully you can do this through Design Pro by trying to adjust the printer or print labels, but the labels will not print). When the printer and PC are synchonized, the led changes to solid green.
If you can synchonize but still can't print, the problem might be the serial cable; not all serial cables will work. You should use a full null model cable (all pins connected except 1 and 8).

SOURCE: TLP 2844 skips over labels
I've had this same problem on the TLP2844. We are using 3 x 1 labels to print shelf barcode labels, it prints a few then skips some labels then prints more. The number printed and skipped are not always the same.
What I did was recalibrate the printer. Turn the printer off. Hold Feed button and turn printer on. The printer will begin feeding labels, looking for the gaps. After the calibration is complete a configuration page will print. Press Feed after to return to normal mode.

I had the same problem. I found out later there was a small piece of label sticking against the sensor, thus blocking the sensor from sensing the end of the label. As soon as I removed the label (open the cover, it is on the top part) it immediately started to work as usual. Hope this helps.

Change orientation in the printer driver.
Open printer & Faxes folder, Double click on printer driver icon,
Zdesigner driver, click on properties, click printing preferences, tick the box "rotate 180%" Click apply & ok.
If driver is Zebra TLP2824, same as above, but hit the + on "Paper output" and turn "rorate 180%" on, then click apply.
