You need to completely uninstall the software and try to reinstall. If the clean install does not work than most likely your system doesn't meet the necessary requirements or the software is not compatible with your system.

See if your ps2 controller hub or port is damaged. If any pin is twisted try to get it straight and if you can't fix it, take the console to a service center.
There are many things that can cause that problem. Try removing it then rensert it again while your NDS is turned off. If that doesn't work, try other game cards if they will work. If other game cards work, then the problem is with the game card that can not be read. I don't know a lot about this console (the Retron 3), but the game sounds like it works fine, try using a different third-party console, I recommend the Retro Entertainment System, it's only around $20 - $25 on eBay, try a different NES game, if it doesn't work, than it's probably the controllers, get replacements, or the batteries are dead.
