When I try to use the "Edit pre set" button, instead of being able to assign a new channel to A3, Only channels pre set on A are available. The manual doesn't even mention the Edit button. Thanks in advance for any help you can provide.

Unlock.....?? What do you mean unlock? The radio is pre programmed with set channels. You would be unable to set your own frequency channels. The FCC has some very stiff penalties for people who use radio frequencies outside of the assigned band and on radios not type accepted for the use. Sorry....
You may have had the receiver set to "Stereo" mode, or Analog instead of "Digital" or "Auto" (automatically switches to digital when it detects a signal) modes. Also, setting it to "Direct" (2-channel from an analog source) could result in loss of center channel. Or your Surround Parameter dial (on the higher-end Denons, at least) could be set to a 2-channel mode. Finally, if your digital inputs are incorrectly assigned (e.g., COAX1 instead of OPT1, or OPT2 instead of OPT1) for that source, the receiver will use the Analog signal which is stereo-only. You could get a center channel via Dolby Pro Logic II but it would be a "matrixed" signal, not the "real" center channel you could get via digital input using Dolby Digital or DTS. (If all else fails, re-check your center channel speaker wire connections. It's very unlikely, but also possible, that either the receiver center channel amp or the speaker has failed.)
Try holding the channel up and volume down buttons together on the front of the set until the set turns off. When you turn it back on it should be normal. You assign the effects basically the exact same way you assigned the channels to inputs, except you press and hold the "MICX4" button and press the "Select" key to the channel (or channels) you want to assign the effects to. Once you assign the effects to the channel, you can edit to the effect you want, and edit the values of the effect.

If I read the manual correctly, you're out of luck without an external tuner such as a cable box. This set was made for the European market and its tuning system isn't fully compatible with that used in the US. There's a "channel swap" function which lets you reassign the channels it finds to whatever channel number you want, and it's described in the manual. I assume that's to allow for the different frequency/channel number assignments in use in all the areas it tries to cover. The set's tuner just may not be able to pick up all the channels on your cable system. As long as it gets US channel 3 or 4 correctly, you can use the set with a cable box or video recorder that properly tunes the cable channels.
Check the instrucitons to verify if the channel table is pre-setable to designate your own numbers.
Most TV sets have an autoprogramming for the channels. Look in the menu for the auto programming and let the set re-program the channels if your set has that option. This feature will re-program the channals, and re-set their defaults.
If the channels are not able to be pre-setable to a custom table, then this indicates that there is a pre-scaling fault in the tuner or in the uPC. In this case, the set will need service.
Re your queries: 1. how to memorize channels? The channels are pre-programmed that a channel number corresponds to a frequency setting. Ex. Ch16 is 156.800 . If for some reasons some channels are missing, perhaps your best course of action is to reset the unit to factory default, that way everything would be back to normal. Master reset is performed by pressing the 16PLUS button while powering the radio. The LCD will display "CL" indicating that the memory has been cleared. If you want to memorize a certain channel/frequency, you can only store it as a replacement for 16PLUS assignment. Ex. Instead of Ch16 as your priority/emergency channel you want say Ch14 (156.700), you can do this by pressing the up/down key till your desired channel; press & hold the 16PLUS key for 3 seconds or until you hear the confirmation beep that your desired channel has been memorized in the memory as the new 16PLUS.
2. Why I cannot transmit? There could be 2 possible reasons: 2.1 You are transmitting on a channel that has been assigned as receive only. Examples are the weather channels, Channels 1,2,3, also 70,75,76, and possibly others. 2.2 Your TX module/circuitry has a problem. More often it is the finals that gets burned.
3. When i use the PTT Channel 16 is automatically set ? The mic has an identical 16PLUS button (similar to the radio's front panel). If you pressed the PTT switch on the side and the radio switches to Ch16 (as if you pressed the 16PLUS button on the front) then you have a wiring problem from the mic to the radio. Possibly a shorted or open connection. It is also possible that you have a break in the small PCB (in some versions) inside the microphone.
