Question about 2006 Harley Davidson XL 1200 C Sportster Custom
If your bike is equipped with a carburetor, it also has a vacuum operated petcock. These things give more problems than I can tell you about. It uses engine vacuum to turn the fuel on and off so you don't have to. The problem is that if the diaphragm gets a hole in it, no fuel goes to the carb even if the tank is full.
I suggest that everyone replace their original petcock with a high quality aftermarket petcock. Pingle makes an excellent manually operated petcock. You'll never have trouble with one of those.

I have the same issue with my 2005 1200 Custom. What make/model of petcock should I get? Do you have a part number that I could use for reference.
